How to know the pixel/percentage values to be set?

I see Nick setting the different values for the properties. He seems to know the exact values to set his webpage right. How do I know what values to set for different properties when I make my own website in the future?

I think most of the time in these videos they are just rebuilding something they've built before, thus he knows exactly what sizes to use. I could be completely wrong, but in the real world it's more trial and error. If there's an exact science to it that I'm missing, I'd love to know it haha
